ABSTRACT

O hambúrguer é um produto cárneo industrializado e potencial veículo de bactérias produtoras ou não de toxinas nas diversas fases de seu processamento, desde sua origem até as fases de transformação, armazenagem, transporte e distribuição para o consumo. Dentro deste contexto, o objetivo do trabalho foi a avaliação do aspecto higienicossanitário dos hambúrgueres bovinos comercializados no Brasil. Foram analisadas 110 amostras de hambúrgueres de carne bovina de 11 marcas comerciais brasileiras segundo a metodologia microbiológica descrita pela Instrução Normativa n° 62 de 26 de agosto de 2003. Todas as 11 marcas comerciais brasileiras apresentaram contagem bacteriana dentro do limite (5x103 UFC/g) para Coliformes a 45°C, enquanto os valores para Staphylococcus coagulase positiva estavam acima dos padrões permitidos (5x103 UFC/g) na marca H (amostras 1 e 7). As marcas B (amostra 5), D (amostra 5), E (amostra 5) e G (amostra 4) também apresentaram contagens acima do estipulado (3x103 UFC/g) para Clostridium sulfito redutor a 46°C e a amostra 6 da marca L foi positiva para Salmonella Infantis.

ABSTRACT

The aim of this study was to evaluate the effect of phytogenic additives and glutamine plus glutamic acid, associated or not, on histomorphometry of bursa of Fabricius and small intestine, oocyst count and lesion scores, and carbon turnover of duodenal mucosa of broiler chickens infected with Eimeria acervulina. A total of 450 male broiler chickens was distributed into a completely randomized design with six treatments and three replications. Treatments consisted of control diet (CD); CD + coccidiosis vaccine; CD + antibiotic performance enhancers and anticoccidial (APE/AC); CD + glutamine and glutamic acid (Gln/Glu); CD + phytogenic additives (PA); CD + Gln/Glu + PA. Birds on treatment CD + vaccine were vaccinated via drinking water at three days of age against coccidiosis. At 16 days of age all birds of all treatments were inoculated orally and individually with 500,000 oocysts of Eimeria acervulina. There was no treatment effect on lesion score in the intestinal epithelium of birds. The smaller number of excreted oocysts was observed in groups of birds fed diets containing APE/AC and PA. Were observed better results of villus height and crypt depth for duodenum and ileum of birds of treatments containing Gln/Glu at 7 days of age, and Gln/Glu and PA at 21 days of age. Higher percentage of cortical area from bursa follicles was observed in birds fed diets supplemented with Gln/Glu and PA at 7, 14 and 21 days of age. Increased turnover of intestinal mucosa was observed in treatments containing Gln/Glu, indicating acceleration in development and regeneration of damaged tissue. Glutamine plus glutamic acid and phytogenic additives can provide improvements to structure, and thus to intestinal function, as well as to better immune response against the infectious challenges. O objetivo deste trabalho foi avaliar o efeito dos aditivos fitogênicos e da glutamina mais ácido glutâmico, associados ou não, sobre a histomorfometria da Bursa de Fabricius e intestino delgado, sobre contagem de oocistos e escores de lesão e sobre o turnover do carbono da mucosa intestinal de frangos de corte experimentalmente infectadas com Eimeria acervulina. Para isso foram utilizados 450 pintos de corte machos distribuídos em delineamento inteiramente casualisado, com seis tratamentos e três repetições. Os tratamentos consistiram de dieta controle (DC); DC + Vacina de coccidiose; DC + antibióticos melhoradores de desempenho e anticoccidiano (AMD/AC); DC + glutamina e ácido glutâmico (Gln/Glu); DC + sditivos fitogênicos (AFs); DC + Gln/Glu + AFs. As aves do tratamento DC + Vacina foram vacinadas via água de bebida, aos três dias de idade, contra coccidiose. Aos 16 dias de idade todas as aves de todos os tratamentos foram inoculadas oralmente e individualmente com 500.000 oocistos de Eimeria acervulina. Não houve efeito dos tratamentos para escore de lesão no epitélio intestinal das aves. O menor número de oocistos excretados foi observado nos grupos de aves alimentadas com dieta contendo AMD/AC e AFs. Foram observados melhores resultados para altura das vilosidades e profundidade das criptas do duodeno e ílio das aves dos tratamentos contendo Gln/Glu, aos 7 dias de idade e Gln/Glu e AFs aos 21 dias de idade. Maior porcentagem de área cortical dos folículos bursais foi observada em aves alimentadas com dieta suplementada com Gln/Glu e AFs aos 7, 14 e 21 dias de idade. Maior turnover da mucosa intestinal foi observada em aves dos tratamentos contendo Gln/Glu, indicando aceleração do desenvolvimento e regeneração do tecido lesado. Glutamina mais ácido glutâmico e aditivos fitogênicos podem oferecer melhorias à estrutura e, consequentemente, à função do intestino, bem como melhores condições para resposta imune frente à desafios infecciosos...

Este trabalho objetivou determinar o turnover do carbono no Pectoralis major, quilha, tíbia e fígado de frangos de corte em seus diferentes períodos de crescimento, utilizando a técnica dos isótopos estáveis. Foram utilizados 216 pintos de corte, machos, Cobb. O delineamento experimental foi inteiramente casualizado e consistiu de sete tratamentos com dias de substituição das dietas distintos: 0; 7; 14; 21; 28; 35; 42 dias de idade. Para mensurar o turnover do carbono nos tecidos em determinado intervalo de tempo, foi utilizado o modelo sigmoidal de regressão de Boltzmann ou função exponencial do tempo. Os valores de meias-vidas foram: 2,5; 2,8; 2,9; 4,9; 5,1; 5,5 e 6,0 dias para músculo peitoral; 2,7; 2,9; 3,1; 4,6; 5,3; 4,1 e 8,0 dias para quilha; 4,4; 3,6; 5,4; 5,8; 4,3; 5,2 e 6,3 dias para tíbia e 1,3; 1,9; 1,4; 1,5; 1,9; 1,7 e 2,1 dias para fígado, nos tratamentos T1, T2, T3, T4, T5, T6 e T7, respectivamente. Pode-se concluir que a tíbia possui metabolismo lento e pode indicar o sinal isotópico das dietas iniciais da criação dos frangos de corte; e o fígado, metabolismo rápido, indicado para refletir a dieta próxima da fase de avaliação.


The aim of this study was to evaluate carbon turnover on muscle tissue Pectoralis major, keel, tibia and liver of poultry at different life stages using the carbon stable isotopes technique. 216 male chick lings (Cobb) were used. The trial was conduced in a totally random distribution consisting on seven treatments with distinct days for diet substitution (0, 7, 14, 21, 28, 35, 42 days old). Either Boltzmann sigmoid regression or the exponential time function were used in order to measure the carbon isotopic turnover in a specific period of time. The half-life were: 2.5; 2.8; 2.9; 4.9; 5.1; 5.5 e 6.0 day on muscle tissue; 2.7; 2.9; 3.1; 4.6; 5.3; 4.1 e 8.0 day on keel; 4.4; 3.6; 5.4; 5.8; 4.3; 5.2 e 6.3 day on tibia and 1.3; 1.9; 1.4; 1.5; 1.9; 1.7 and 2.1 day on liver, for to treatments T1, T2, T3, T4, T5, T6 e T7 respectively. According to the results of this study, it is possible to conclude that the tibia has slowest metabolism and may indicate the isotopic signature of initial diets of poultry culture. The fast metabolism tissue, liver, is appropriated to reflect the diets fed at closer period of evaluation.

Background Environmental devastation threatens the survival of many species, including venomous snakes such as the South American rattlesnake Crotalus durissus terrificus. This observation is based on the decrease of snakes collected and donated to Brazilian research institutes. Nevertheless, some individuals have managed to survive and procreate. The question is how these snakes are adapting in these new environmental conditions.Methods To answer it, the carbon-13 level of rattlesnakes and their feed (either laboratory or wild mice) was evaluated by isotope-ratio mass spectrometry. Thus, rattle segments from 16 adults and 15 offspring of captive snakes, and of three wild newborn C. d. terrificus were evaluated as well as 17 Mus musculus mice captured in traps, four live feeder mice and the ration offered to mice at animal houses.Results The isotopic exchange time of the captive adult snakes (n = 16) varied between 33 and 37 months and of captive-born animals (n = 15), until reaching a plateau of equilibrium, varied from 18 to 24 months. Regarding the captured Mus musculus (n = 17), 88.23% (n = 15) were from a C4 environment. Of the six rattle rings from offspring of captured C. d. terrificus, five were from a C4environment, whereas of the 170 rattle rings studied, 60% originated from a C3 environment and 40% from a C4. The same carbon-13 values were found in captive snakes.Conclusions Based on the present results, it can be inferred that most C. d. terrificus snakes (60%) fed animals from a C3environment; birds consist of an alimentary alternative for snakes, as well as rodents, small reptiles and amphibians; different venom compositions among snakes from the same region may be related to the food type; the primary rattle of offspring reflects the maternal diet during gestation; and, finally, the different rattle rings indicate the alimentary history of these animals.(AU)

The present study investigates the δ13C and δ15N isotopic composition in frozen samples (control), samples in alcohol and in formaldehyde of Plagioscion squamosissimus and Hypophthalmus edentatus. From each individual we extracted a strip of muscle from the region above the lateral line, in the dorsal fin base, that was divided into three equal parts, each one was submitted to one type of treatment: freeze ­ control group (-15oC), conservation in alcohol 70% and fixation in formaldehyde 4%. Samples were kept under those treatments for 30 days, washed and submerged in distilled water for 4 hours. Afterwards, they were dried up in air oven at 60oC for 48 hours and macerated until the obtaining of a fine powder. A significant difference was found in isotopic values of carbon and nitrogen, between the control and the samples in alcohol and formaldehyde, except for δ13C from the H. edentatus samples in formaldehyde. The carbon isotopic values of samples in alcohol were mostly enriched compared to control, whereas the samples in formaldehyde presented depleted values in relation to the control. The nitrogen isotopic values for both samples preserved in alcohol and formaldehyde were enriched when compared to the values of frozen samples, independently of used preservatives. Therefore, the isotopic correction should be accomplished according to the isotope and preservative employed for species of freshwater fish.


O presente estudo investiga a composição isotópica de δ13C e δ15N entre as amostras congeladas (controle), em álcool e em formol de Plagioscion squamosissimus e Hypophthalmus edentatus. De cada indivíduo foi extraída uma faixa de músculo localizada na região acima da linha lateral, na base da nadadeira dorsal, a qual foi subdividida em três partes iguais, sendo cada uma delas submetida a um tipo de tratamento: congelamento - grupo control (-15oC), conservação em álcool 70% e fixação em formol 4%. As amostras foram mantidas nos referidos tratamentos por 30 dias, enxaguadas e submersas em água destilada por 4h. Em seguida, foram secas em estufa de ventilação a 60oC por 48h e maceradas até a obtenção de pó. Identificou-se diferença significativa nos valores isotópicos de carbono e de nitrogênio, entre o controle e as amostras de álcool e as de formol, com exceção do δ13C das amostras mantidas em formol de H. edentatus. Constatou-se que os valores isotópicos de carbono das amostras conservadas em álcool foram, na sua maioria, enriquecidos quando comparados com as controle, ao passo que, as amostras em formol tiveram os valores deplecionados em relação ao controle. Os valores isotópicos de nitrogênio, tanto para as amostras conservadas em álcool, quanto em formol, foram enriquecidos quando comparados aos valores daquelas congeladas, sendo estes independentes dos conservantes utilizados. Assim, a correção isotópica deve ser realizada de acordo com o isótopo e o conservante empregado para espécies de peixe de água doce.

ABSTRACT

O objetivo deste trabalho foi avaliar o efeito da dieta suplementada com nucleotídeos sobre taxa de turnover da mucosa intestinal de frangos antes e após lesões causadas por coccidiose por meio de análise isotópica, utilizando a variação do carbono-13 em plantas C3 e C4. Foram utilizados 264 pintos submetidos a dois tratamentos: dieta controle e dieta suplementada com 0,07 por cento nucleotídeos. Os pintos possuíam sinais isotópicos em seus tecidos semelhantes ao de ditas C4. Após o alojamento, para avaliar taxa de turnover da mucosa, as aves receberam dietas predominantemente C3. Com 16 dias de idade, 50 por cento das aves de cada tratamento foram inoculadas com oocistos de Eimeria acervulina e, a partir de 21 dias, para avaliar a taxa de turnover após desafio, as aves passaram a receber dieta predominantemente C4. Amostras de mucosa foram analisadas isotopicamente por espectrometria de massa. Na fase inicial, a suplementação com nucleotídeos propiciou aceleração na troca do carbono na mucosa, com meia-vida de 1,06 e 1,01 dias para dieta controle e com nucleotídeos, respectivamente, acelerando o crescimento intestinal. Na fase de 21 a 42 dias de idade, para os grupos não desafiados com coccidiose, as meias-vidas do carbono foram 1,81 e 1,80 dias para dieta controle e com nucleotídeos, respectivamente, não havendo influência dos tratamentos. Porém, nos grupos desafiados, as meias-vidas foram 1,01 dias no grupo controle e 0,75 dias no que recebeu dieta com nucleotídeos, indicando maior turnover da mucosa nesse último grupo. A adição de nucleotídeos promove aceleração no processo de renovação da mucosa intestinal e na regeneração após danos causados por coccidiose.


The objective was to evaluate the effect of nucleotides supplementation diet on the intestinal mucosa turnover rate of broilers before and after injury caused by coccidiosis, through isotopic analysis, using the carbon-13 variation in C³ and C4 plants. It was used 264 chicks under two treatments: control diet and diet supplemented with 0.07 percent nucleotides. The chicks had isotopic signals in their tissues similar to C4 diet. After housing, to assess mucosal turnover rate, the birds were fed with a predominantly C3 diet. At 16 days of age, 50 percent of birds from each treatment were inoculated with Eimeria acervulina oocysts and starting from 21 days, to assess the mucosa turnover rate after challenge, the birds received predominantly C4 diet. Samples of intestinal mucosa were isotopically analyzed by mass spectrometry. In the initial phase, supplementation with nucleotides led acceleration in carbon trading in the mucosa, with half-lives of 1.06 and 1.01 days for control diet and with nucleotides, respectively, accelerating intestinal growth. During the 21 to 42 days of age, for not coccidiosis challenged groups, the carbon half-lives were 1.81 and 1.80 days for control diet and with nucleotides respectively, without any influence of the treatments. However, in the challenged groups, the half-lives were 1.01 days in control group and 0.75 days in group receiving nucleotides diet supplementetion, indicating a higher mucosa turnover rate in the latter group. The addition of nucleotides promotes acceleration in the cell renewal process of intestinal mucosa and in regeneration after damage caused by coccidiosis.

ABSTRACT

No presente trabalho, determinaram-se as trocas gasosas de folhas de figueira 'Roxo de valinhos' e o ciclo fotossintético por meio da relação isotópica 12C/13C. Essas medidas foram realizadas sempre na região mediana das folhas, completamente expandidas e totalmente expostas à radiação solar, no período das 09h00min às 10h30min. As folhas fotossinteticamente ativas da figueira apresentaram área foliar em torno de 160cm², com uma assimilação de 14,38µmol m-2 s-1 de CO2, cujos valores isotópicos médios no ramo 1 e no 2 são -28,98±0,69‰ e -29,28±0,85‰, respectivamente. Com base nos valores da fotossíntese máxima e na discriminação isotópica do 13C, evidenciou-se que a figueira pode ser considerada uma planta do ciclo fotossintético C3.


In the present research, it was determined the gas exchange of the 'Roxo de Valinhos' fig tree and the cycle photosynthetic through the isotopic relation 12C/13C. These measures were always carried in the average region of the leaves, completely expanded, entirely displayed to the solar radiation, in the period from 09h00min to 10h30min. The sheets photosynthetic active leaf area had around 160cm², with 14.38 -2 s-1 assimilation CO2, and mean isotopic values in the branch 1 and 2 of -28.98±0.69‰ and -29.28±0,85‰, respectively. Based in the values of the maximum photosynthesis and in the discrimination isotopic of the 13C, the fig tree can be considered a plant which belongs to photosynthetic C3 cycle.

ABSTRACT

Neste trabalho, objetivou-se analisar isotopicamente méis comercializados nas regiões Sul e Sudeste do Brasil, para a detecção de fraude. Foram colhidas amostras comerciais com registro no Serviço de Inspeção Federal, Estadual ou Municipal. As amostras foram submetidas à combustão no Analisador Elementar EA 1108 CHN e analisadas no espectrômetro de massas de razão isotópica DELTA-S (Finningan Mat). Os valores isotópicos (δ13C) dos méis in natura foram comparados aos de suas respectivas proteínas (padrão interno). Foram consideradas adulteradas as amostras cuja diferença entre o valor isotópico da proteína e do mel foi igual ou inferior a -1‰. As amostras consideradas adulteradas pela análise isotópica foram submetidas a testes químicos qualitativos que não foram capazes de indicar adulteração para algumas delas. Das 61 amostras analisadas, 18,0 por cento encontram-se adulteradas, sendo 11,5 por cento na Região Sudeste e 6,5 por cento na Região Sul. Ao contrário dos testes químicos, a análise isotópica mostrou-se eficaz em identificar e quantificar a adulteração de méis comerciais.


The aim of this study was the isotopic evaluation of honey traded in the Southern and Southeastern Brazilian regions, to detect fraud. Commercial samples, registered in the municipal, State or Federal Inspection Service, were collected and submitted to combustion in the EA 1108 CHN Elemental Analyzer and analyzed in the DELTA-S (Finningan Mat.) isotope ratio mass spectrometer. The isotopic values (δ13C) of in natura honey were compared to their respective proteins (internal standard). Samples whose difference between the isotopic value of protein and honey was equal or inferior to -1‰ were considered adulterated. The samples considered adulterated were submitted to qualitative chemical tests which were unable to show adulteration for some of them. Among the 61 samples analyzed, 18.0 percent were adulterated; 11.5 percent in the Southeastern and 6.5 percent in the Southern region. Unlike chemical tests, isotopic analysis has shown to be efficient to identify and quantify adulteration in commercial honey.

Oobjetivo deste trabalho foi determinar a quantidade de malte e de adjunto presentes nas cervejas comerciais brasileiras do tipo Pilsen e detectar possível adulteração em sua composição, tomando por base a legislação brasileira. Para isso, foi utilizada a metodologia de análise isotópica utilizando os isótopos estáveis dos elementos carbono e nitrogênio. Foram analisadas 161 amostras de cervejas provenientes de dezessete estados do Brasil. Concluiu-se que 95,6 por cento utilizaram malte e adjunto cervejeiro (derivados de milho ou açúcar de cana: 91,3 por cento e derivados de arroz: 4,3 por cento) em sua formulação e outros 4,3 por cento eram cervejas "puro malte". Das amostras analisadas, 3,7 por cento eram cervejas com menos de 50 por cento de malte em sua formulação (adulteradas), 28,6 por cento delas estavam na faixa de incerteza, para qualquer tipo de adjunto e 67,7 por cento apresentaram malte dentro do limite legal estabelecido.


The aim of this study was to determine the amount of malt and adjunct in Pilsen Brazilian commercial beer, and to detect likely adulteration in its composition, based on the Brazilian legislation. The methodology applied was the isotopic analysis which used carbon and nitrogen stable isotopes. One hundred sixty-one beer samples from seventeen Brazilian states were analyzed. It was concluded that 95.6 percent used malt and adjunct (corn or sugarcane derived: 91.3 percent and rice derived: 4.3 percent) in their formula and 4.3 percent were "pure malt" beer. Among the analyzed samples 3.7 percent were beers with less than 50 percent of malt in their formula (adulterated), 28.6 percent of them were in doubtful level for any adjunct and 67.7 percent contained malt in the legal established limit.
